Is there a way to create a hyperlink within the comments section of an issue that leads directly to an internal-network-drive folder?

If your browsers support the "file://" prefix and recognise network drive names with it, or to open a file manager when they see it, then yes.  You can simply qualify the whole resource string for the folder.
