I have 2 agents(developers) that need to be restricted from others agents to be assigned to issues. In other words, only admins would be allowed to assign them issues. Currently, any desk agent can assign issues to each other on this project.

Unfortunately, this cannot be done to restrict assigning issues to you two users (agent) to be exclusively done by Admins. This is because the "Assignable User" + "Assign Issues" are controlled at the project level.
The only way you may want to try is removing the ability of "Assign Issue" right against those two specific users. This means that you will need to update the "Assign Issue" right accordingly and those two users (agents) can no longer have the ability to assign issues in your project.

Based on your update
The idea is that the 2 agents are specialists so we don't want them to be working or assigned to issues of low priority and the managers (admins) are the only ones able to assign them work/issues on that project.
The best option I can think of is for you to use Automation for Jira and setup the rule to trigger off by Issue Created event, and afterward add an conditional check for all "low priority" issues then check for issue assignee value. If assignee is your specialist, then override the issue assignee to someone else.

Should these 2 agents be barred from being assigned to all issues across projects, specific issuetypes, or just occasional issues?
The reason I ask is that you may be able to achieve what you need with Issue Security Schemes.
You could create 2x Security levels, 1 for the 2x agents that shouldn't see stuff, and 1 for everyone else.
So that way the 2x agents could still be assigned issues, but they could only see the contents and interact with it if the issue security level matches the level that they belong to.

The 2 agents (developers) are active on our software projects, but occasionally they would need to interact or be assigned to issues on the service desk project. We just don't want other agents to automatically assigned them issues just because they know that the issues would fall into their domain.
