I want to create a report that would show the tickets that haven't had any Comments under Activity updates for let's say 2 days. Is this possible?

Welcome to the community.
Jira does not have a native method for searching based on dates of Comments.
Are you open to using a third party app? If so there are several that extend the searching capabilities of Jira to allow you to search on additional criteria/fields.
If you are not open to using a third party app, then the work around would be to create a custom field to track the last date a comment was added, and use an Automation Rule to update the field each time a comment is added. Then search against that field.
