What does it mean when it says a transition can't be configured when the workflow is in draft mode?

It means that, as the workflow you are currently editing is active and in use by at least one project, then you have to make a copy of said workflow, do the changes you couldn't do when receiving that message, and finally set that modified workflow to substitute its previous version.

Exactly what it says - there are some things you can't do with workflows when in draft mode.
