Yep. That's all handled by the notification scheme of your project, but make sure you created a mail handler beforehand. It's all explained in the documentation.
You don't need a mail handler to get notifications to go out of JIRA - they're for processing incoming email.
You do need to think about what you want to do if a user clicks "reply" though, and an email handler is a really good option.
