Do I need to set up the "Server Setting -> Subversion client" if I access a Subversion repository using the protocol file://
Also do I need to specify a usename/password using the protocol file://

You do not need to specify a username and password if using the file protocol.
The specification of subversion client is optional but if you specify one, it will be used for all subversion queries including using the file:// protocol. Fisheye bundles the SVNKit Java SVN library, and this will be used to access Subversion if you do not specify a client.
