Using code, no, because you can't upload plugins into OnDemand (and I don't think Connect is going to be that powerful)
Also, what does "from Jira to Confluence" mean? Where are you trying to update and what effect should it have on the other system? Even then, the answer is probably "depends entirely on what user management system you are using"
You can have JIRA do this for you by using the JIRA User Server.
Just do this to connect JIRA and Confluence:
After that, you can just go to the Confluence Admin page and under User Directories press "Synchronize".

If you are using OnDemand, you anyway have centralized user management. Are both your JIRA and Confluence hosted in OnDemand?

i can able to perform this one manually in the UI on Ondemand JIRA but i need the detail of how can i achieve through the REST API. By using REST API how can i activate and deactivate the user in confluence based on the JIRA user. It is possible to perform this one.
