when transition happened some custom filed values should update automatically?

After doing transition custom fields value should be updated to perticular value and it it should not be editable in the transition or view screen.

how we can achive this in the workflows postfunction?

Set the fields with post-functions, then remove them from the transition and edit screens. The in-line edit respects the "edit" screen, so you won't be able to inline edit them in the view screen.
